Intermediate Filaments and the Regulation of Cell Motility
Cellular and Molecular Biology of Intermediate Filaments
IFs Summary (LODISH BK. M.B) The association of intermediate filaments with the nuclear and plasma membranes suggests that their principal function is structural. In epithelium, for instance, intermediate filaments provide mechanical support for the plasma membrane where it comes into contact with other cells or with the extracellular matrix. They are distinguished from "thin filaments" by their size (8-10 nm) and the fact that thin filaments are obviously motile. “Intermediate filaments” is actually a generic name for a family of proteins (grouped into 6 classes based on sequence and biochemical structure) that serve similar functions in protecting and shaping the cell or its components. Interestingly, they can even be found inside the nucleus. As we just stated, intermediate filaments are cytoskeletal components of cells that are mid-sized of the three cytoskeletal fibers. Think of them as the small pickup truck of these fibers.

The latter is achieved by anchoring the chromatin to the nuclear lamina that lines the inner part of the nuclear membrane. Intermediate filaments are about 10 nm in diameter with wavy profiles in electron micrographs of thin sections of cells (Fig. 35-1B) or after negative staining isolated filaments . In some cases, the N- or C-terminal domains project from the surface ( Fig. 35-3C ). Intermediate filaments organize the internal 3D structure of the cell, anchoring all the organelles in place and serving as structural components of the nuclear lamina and sarcomeres. They also participate in some cell-cell and cell-matrix junctions.

They have a coiled structure that provides mechanical support to the cell as they stretch from the nucleus throughout the cytoplasm. Cells that are subject to mechanical stress, such as hair and skin cells, contain a higher amount of intermediate filaments compared to other cell types. 1989-12-01 In general, intermediate filaments serve as structural elements, helping cells maintain their shape and integrity.

Another type of intermediate filament, the nuclear lamins (5) are responsible for maintainingthe structure of the nucleus. 5. EGF in cell culture media results in partial de-differentiation of astrocytes and therefore should be used with caution. Keywords: intermediate filament (nanofilament) proteins, nestin, neural stem/progenitor cells, astrocytes, GFAP, vimentin, HB-EGF ISBN: 978-91-628-9047-6 Intermediate filaments have an average diameter of 10 nanometers, which is between that of 7 nm actin (microfilaments), and that of 25 nm microtubules, although they were initially designated ‘intermediate’ because their average diameter is between those of narrower microfilaments (actin) and wider myosin filaments found in muscle cells. Intermediate filaments anchor the nucleus and position it within the cell, and they give the cell its elastic properties and its ability to withstand tension. Microtubules ( MT) are long, hollow filaments made of globular monomeric proteins, "alpha and beta-tubulin.
